A visual colorimetric dual readout bioassay for determination of pesticide residues in drinking water

  Abstract  
  A simple and practical visual detection method has been developed and described for the quantitative in-field analysis of organophosphates (OPs) and carbamate (CB) residues in drinking water using microplate reader. The method utilizes a sensitive alkaline phosphatase (ALP from calf intestine) immobilized silica (SiO2) micro-column. The inhibition is visualized on the micro-column using 0.09 U ALP and 0.143 mM para nitrophenylphosphate (pNPP) in 60 µL solution. The inhibition by OPs and CB was studied in the range 0.01-500 µg L-1 and was visually distinct. Inhibition of ALP activity was quantified for methyl paraoxon (MPOx), ethyl paraoxon (EPOx), methyl parathion (MP) and carbofuran (CF) in drinking water. No interferences were observed when the assay was tested in the presence of metal ions Hg2+, Cd2+ and Pb2+ at low level. The method can easily quantitate residual pesticide concentration as low as 0.1 µg L-1 with relative standard deviation (RSD) of 1.6%.
