Simultaneous estimation of amlodipine, hydrochlorothiazide and losartan by RP-HPLC

  Abstract  
  A reverse phase high performance liquid chromatography method has been developed for simultaneous estimation of Amlodipine, Hydrochlorothiazide and Losartan. The separation of these three drugs was achieved on a Phenyl column (250mm X 4.6mm, 5µm) as stationary phase with a mobile phase consisting of Acetonitrile : Buffer (6.64g sodium di-hydrogen phosphate + 0.84g sodium perchlorate in 1000 mL distilled water, pH-2.0 with orthophosphoric acid and 3ml of triethyleneamine) in the volume ratio of 45:55 at a flow rate of 1.0 mL/min and UV detection at 254nm. The retention times were observed to be 4.3, 3.3 and 6.2 minutes for Amlodipine, Hydrochlorothiazide and Losartan respectively. The method was validated for linearity, recovery, limit of detection, limit of quantification, accuracy and precision. The method was successfully applied for analysis of tablet dosage form containing Amlodipine, Hydrochlorothiazide and Losartan.
