Determination of pKa values of opipramol in acetonitrile-water binary mixtures by using chromatographic and spectrophotometric methods

  Abstract  
  The chromatographic pKa value was determined based on the retention behavior of opipramol at different pH conditions. The effect of the mobile phase composition on this ionization constant was studied by measuring the pKa at different acetonitrile-water mixtures, ranging 35-45% (v/v), using LC method. In doing so, the pH values were measured in the hydroorganic mixture, which was used as the mobile phase, instead of in water, taking into account the effect of activity coefficients at 25 oC. An octadecylsilica Nucleosil C18 column (150 mm × 4.6 mm, 5 µm) was used for all the determinations. The reliability of the LC methodology was investigated statistically. The pKa values of opipramol were also determined by using spectroscopic method at different percentages of acetonitrile water binary mixtures, ranging 20-40% (v/v). Aqueous pKa values of opipramol were calculated by mole fraction and Yasuda- Shedlovsky equations. Moreover, estimation of aqueous pKa values can also be predicted by computational methods on the basis of molecular structure. Results obtained from pKa values are compared with these programs and literature values.
